Ideas in this flowchart

  • Clinical activity and future evaluation of dual PD-(L)1 and VEGF inhibition in NSCLC
  • Confirmed ORR was encouraging across doses and histologies
  • The cORR was 57.1% in nonsquamous NSCLC
  • The cORR was 68.4% in squamous NSCLC
  • cORR was similar across PD-L1 expression levels
  • Activity was observed in patients with very low PD-L1 expression, including TPS below 1%
  • This subgroup generally has poor response to immune checkpoint inhibition
  • ctDNA levels declined across all evaluable patients
  • ctDNA was measured with the FoundationOne Liquid CDx 324-gene panel
  • ctDNA clearance rates were similar for both histologies
  • The findings add to enthusiasm for dual PD-(L)1 and VEGF inhibition as a next-generation immunotherapy strategy
  • Activity was seen across PD-L1 expression levels, including PD-L1-negative tumors
  • PD-L1-negative tumors have a high unmet need
  • Response rate is an important early signal
  • The key question is whether the approach improves progression-free survival, overall survival, and tolerability
  • These outcomes must be compared with current pembrolizumab-based chemoimmunotherapy

"Confirmed ORR (cORR) was “encouraging” across doses and histologies, Dr Peters said, at 57.1% for nonsquamous NSCLC patients and 68.4% for squamous NSCLC. cORR was similar across PD-L1 levels, including the 21 patients with very low PD-L1 (TPS <1%) expression, a subgroup with generally poor response to immune checkpoint inhibition, Dr Peters noted.\n\nCirculating tumor DNA (ctDNA) levels measured with the FoundationOne Liquid CDx 324-gene panel declined across all evaluable patients, with similar ctDNA clearance rates for both histologies, she said.\n\n“These data are interesting and add to the growing enthusiasm around dual PD-(L)1 and VEGF inhibition as a next-generation immunotherapy strategy in NSCLC,” said Dr Schoenfeld. “Activity was seen across PD-L1 expression levels, including PD-L1-negative tumors, where unmet need remains high.”\n\nResponse rate is an important early signal, Dr Schoenfeld noted, but “the key question moving forward will be whether this approach improves progression-free survival, overall survival, and tolerability compared with current pembrolizumab-based chemoimmunotherapy.”"
