  • Phase 2b trial comparing T+D+CT with P+CT in patients
  • The trial randomly assigned 84 patients: 41 to T+D+CT and 43 to P+CT.
  • T+D+CT arm: 41 patients
  • Patients received 75 mg tremelimumab, 1500 mg durvalumab, and pemetrexed-platinum chemotherapy every 3 weeks for 4 cycles.
  • Maintenance treatment consisted of durvalumab and pemetrexed every 4 weeks until disease progression, with additional tremelimumab at week 16 and optionally at month 24.
  • P+CT arm: 43 patients
  • Patients received 200 mg pembrolizumab plus chemotherapy every 3 weeks until disease progression, for up to 24 months.
  • Confirmed objective response rate was 39.0% with T+D+CT and 34.9% with P+CT.
  • Median duration of response was not reached with T+D+CT and was 6.4 months with P+CT.
  • At 6 months, 100% of responders in the T+D+CT arm remained in response, compared with 58.3% in the P+CT arm.
  • The reported findings came from the presentation abstract for trial NCT06008093.

"In this phase 2b trial (NCT06008093), researchers randomly assigned 84 patients to receive T+D+CT (n=41) or P+CT (n=43). According to the presentation abstract, patients in the first group received 75 mg tremelimumab, 1500 mg durvalumab and pemetrexed-platinum chemotherapy every 3 weeks for 4 cycles, then maintenance durvalumab and pemetrexed every 4 weeks until disease progression; additional doses of tremelimumab were given at week 16 and optionally at month 24. The control group received 200 mg pembrolizumab plus chemotherapy every 3 weeks until disease progression, for up to 24 months. The confirmed ORR was 39.0% with T+D+CT and 34.9% with P+CT. The median duration of response was not reached and 6.4 months, respectively. One hundred percent of responders in the T+D+CT arm were still in response at 6 months, compared to 58.3% of patients in the P+CT group."
