Ideas in this flowchart

  • mAbs were cloned and characterized for binding and cross-recognition of WT S protein.
  • Strong plasma ADCC activity supported the conduct of mAb cloning.
  • Memory B cells recognizing WT S protein were individually sorted.
  • The sorted CD19+ CD27+ IgG+ Spike+ cells were used for B cell receptor cloning.
  • ELISA was used to evaluate the binding of the resultant mAbs to WT S protein.
  • Several resultant mAbs bound WT S protein strongly, with EC50 values below sub-nM levels.
  • Different S protein domains were used to identify the binding regions of the mAbs.
  • The mAbs did not restrict binding to specific WT S protein subunits, including NTD, RBD, or S2.
  • Several mAbs also showed cross-recognition activity.

"showed strong plasma ADCC activity ( Fig. 1b ) to conduct mAb cloning. Memory B cells recognizing WT S protein (CD19 + CD27 + IgG + Spike + ) were sorted individually ( Fig. 3b ) for B cell receptor cloning. The ELISA results revealed that several resultant mAbs bind strongly with WT S prote in with EC 50 lower than sub - nM levels ( Fig. 3c and Supplementary Fig. 3a ). We used different domains of the S protein to identify the binding region of these mAbs ( Supplementary Fig. 3b ). The results revealed that they do not restrict binding to certain subunits of WT S protein, such as NTD, RBD, and S2 regions ( Fig. 3c and Supplementary Fig. 3b ). Several of these mAbs also showed cross - recognition activity ( Fig. 3c )."
