ASCO 2026: Lung Cancer Treatment Options, New Studies - Cancer Therapy Advisor / CHRYSALIS-2: First-Line Amivantamab Plus Lazertinib

"First-line amivantamab plus lazertinib (ami-laz) more than doubled median OS compared to historical outcomes for afatinib, at 41 months compared to 19.4 months, and 55% of patients with EGFR-mutated advanced NSCLC were alive at 3 years, reported study coauthor Joel W. Neal, MD, PhD, of the Stanford Cancer Institute at Stanford University in California.1\n\n“We believe this is a clinically meaningful median overall survival of almost three and a half years among 49 patients on this study,” Dr Neal told Cancer Therapy Advisor. “The responses were durable regardless of demographics, tumor mutations, or other disease characteristics.”\n\nThe safety profile was consistent with prior reports, he noted.\n\n“With a follow-up of over 31 months across all of the population, we observed an overall survival median of 41 months, with 85% of patients still alive at 1 year, 72% at 2 years, and 55% at 3 years,” Dr Neal said. “We looked at subgroup analyses to see whether there were patterns with respect to age, race (Asian or not), or presence of brain metastases at baseline — and didn’t see that any of these predicted longer time on treatment.”\n\nLooking at real-world data from 2014 to 2024 from patients with similar eligibility as Cohort C of CHRYSALIS-2 as an OS comparator, the researchers found that physician-selected EGFR tyrosine kinase inhibitors were associated with an OS of only 15.2 months, Dr Neal noted."
