ASCO 2026: Lung Cancer Treatment Options, New Studies - Cancer Therapy Advisor / CROWN: 7-Year Update for Lorlatinib
"Lorlatinib’s safety was consistent with earlier analyses, with no new signals during follow-up. Most grade 3 or 4 adverse events since the prior 5-year analysis were increased lipid values, Dr Mok reported, but despite higher any-grade hyperlipidemia rates with lorlatinib (71% vs 6%), there was no increase in cardiovascular adverse events compared to patients in the crizotinib study group. Cognitive and mood effects of any grade were reported in 30% and 21% of patients taking lorlatinib, respectively, compared to 7% and 7% of those on crizotinib.\n\nLong-term efficacy in the lorlatinib group was similar between patients who underwent dose reductions within 26 weeks and those who did not, Dr Mok noted."
